چکیده

l-Fucose isomerase (l-FucI)-mediated isomerization is a promising biotechnological approach for synthesizing various rare sugars of industrial significance, including l-fucose. Extremozymes that can retain their functional conformation under extreme conditions, such as high temperature and salinity, offer favorable applications in bioprocesses accompany harsh conditions. To date, only one thermophilic l-FucI has been characterized l-fucose synthesis. Here, we report from Halothermothrix orenii (HoFucI) which exhibits both halophilic properties. When evaluated biochemical HoFucI exhibited optimal activities at 50–60 °C pH 7 with 0.5–1 M NaCl the presence 1 mM Mn2+ cofactor. The results obtained here show unique feature polyextremozyme, facilitates production using this enzyme.

An epimerase-reductase in L-fucose synthesis.

The first committed enzyme in GDP-L-fucose formation from GDP-D-mannose is GDP-D-mannose 4,6-dehydratase, which forms GDP-4-keto-6-deoxy-D-mannose. The uncertain enzymatic steps beyond this point were examined in this study. Assays were developed for the epimerase and reductase activities which the putative pathway would predict. A protein was isolated exhibiting homogeneity by several criteria...

The Metabolism of L - Fucose

The isomerization of L-fucose to L-fuculose was observed by Green and Cohen (2) in extracts obtained from Escherichia coli grown on L-fucose. With the same organism and similar conditions, Huang and Miller (3) suggested that such extracts catalyzed the phosphorylation of both L-fucose and L-fuculose to form the corresponding sugar l-phosphates. The present report describes the partial purificat...

A bifunctional enzyme with L-fucokinase and GDP-L-fucose pyrophosphorylase activities salvages free L-fucose in Arabidopsis.

Monomeric sugars generated during the metabolism of polysaccharides, glycoproteins, and glycolipids are imported to the cytoplasm and converted to respective nucleotide sugars via monosaccharide 1-phosphates, to be reutilized as activated sugars. Because L-fucose (L-Fuc) is activated mainly in the form of GDP derivatives in seed plants, the salvage reactions for L-Fuc are expected to be indepen...

Bacterial dissimilation of L-fucose and L-rhamnose.

Eagon, R. G. (University of Georgia, Athens). Bacterial dissimilation of l-fucose and l-rhamnose. J. Bacteriol. 82:548-550. 1961.-Of 33 microorganisms screened for the ability to utilize the methylpentoses, l-fucose and l-rhamnose, 18 species utilized one or both methylpentoses. The majority of the bacterial species capable of utilizing these methylpentoses did so by isomerization of the methyl...

Metabolite Control of L - Fucose Utilization 8361

Thyroid fucokinase is responsive to a number of metabolites which might serve in a regulatory capacity. In addition to inhibition by ADP and stimulation by GMP, fucokinase responds selectively to a series of nucleotide sugars. Of those studied, only guanine nucleotide sugars moderate the activity of the enzyme. GDP-a-D-mannose, GDP-a-D-glucose, GDP-cY-D-rhamnose, and GDP-cu-L-rhamnose all stimu...
